A common factor in media getting lost in the first place is a lack of interest. Regardless of who cares about it, it's still considered lost. What if somebody sees an article on a lost dub and they happen to have footage from it? Then we will have found something, and that's always a victory in my book.

This revelation raises the question if the trailer you were looking for was actually a fan-made modification for a Grand Theft Auto game that was released on the PlayStation 3 or Xbox 360. As far as I know, the Grand Theft Auto series has one of the most popular and dedicated fan modding communities that creates new content and makes re-skins of existing characters. The same can be said for the Super Mario Bros. franchise (which now has its own official level creator called Super Mario Maker for the Wii U and Nintendo 3DS), The Sims series (which allowed custom content from the start), and even Jurassic Park: Trespasser, a PC game that barely sold 50,000 copies and was noted for being too admittedly ambitious for the time and having several bugs and glitches, have dedicated modding communities willing to create new content and levels for the games they love and maybe fix up some bugs the original game suffered from.
Back on track, if you said that the Super Mario Bros. characters looked like realistic humans in the Grand Theft Auto series, I suggest looking up on YouTube for a specific game in the series and search for fan-made modifications for that said game, I'm sure you'll find plenty of mods of the game in question and maybe find one similar to your own description.
I guess I also should have clarified that I didn't mean that I thought the game had any relation to Grand Theft Auto. I said the characters looked as realistic as humans in video games looked at the time, and Grand Theft Auto was the first game to come to mind. When I say "realistic", I mean like on the level of Grand Theft Auto, Hitman, Call of Duty, etc.. This couldn't be a game from any of those franchises as it was a fighting game.

Are you sure it is not an official trailer for a real Mario game? If so, I think that Mario Striker's Charged (a 2007 Wii sequel to the 2005 Nintendo GameCube game Super Mario Strikers) fits the closest to what you have described because it is one of the few games in the series to have receive an E10+ rating from the ESRB but it's not bloody but more realistic than other the Super Mario Bros. games.
Also, from what I read about the game, the playable team captains were Mario, Luigi, Peach, Daisy, Yoshi, Wario, Waluigi, Donkey Kong, Diddy Kong, Bowser, Bowser Jr. and Petey Piranha. The game also featured sidekicks for your teams including Birdo, Toad, Boo, Koopa Troopas, Hammer Bros., Dry Bones, Monty Moles and Shy Guys.
